Detecting Doxorubicin Cardiotoxicit in Rats Morphological and Hemodynamic Findings

چکیده
Background. The therapeutic value of doxorubicin (DOX) is limited by its cardiotoxicity, which is dose dependent. To improve the detection of such myocardial damage, this study was designed to determine whether the "'In antimyosin antibody Fab could serve as a marker for cardiotoxicity in treated versus control rats on the basis of comparative morphological and hemodynamic findings. Methods and Results. DOX was administered by intravenous injection to rats at a dose of 3 mg. kg' week` for 5 weeks. Three weeks after the final injection, an intravenous dose of "'In antimyosin, 740 kBq (20 pCi), was administered, and tissue distribution of the radioisotope (percent kilogram dose per gram) was assessed in 48 hours. Myocardial uptake of radioactivity by both ventzicles was more prominent in the DOX-treated rats than in control rats (p<0.001). The heart-to-blood and heart-to-lung uptake ratios were markedly higher in the treated rats than in controls (p<0.001). As the severnty of the myocardial damage increased, there was a progressive increase in myocardial uptake. There

Therapeutic effect of valsartan against doxorubicin-induced renal toxicity in rats
Objective(s): Doxorubicin (DXR)-induces glomerular atrophy and fibrosis in rat kidneys. The objective of the current study was to investigate the protective effects of valsartan on DXR-induced glomerular toxicity and its mechanisms of actions in rats. Materials and Methods: Male Sprague-Dawley (SD) rats were divided into four groups, and each group contains ten rats. First group was control and...
متن کاملEffect of Green Tea Extract on Doxorubicin Induced Cardiovascular Abnormalities: Antioxidant Action
Doxorubicin (DOX) induces oxidative stress leading to cardiovascular abnormalities. Green tea extract (GTE) is reported to possess antioxidant activity mainly by means of its polyphenolic constituent, catechins. Our study was aimed to find out the effect of GTE (100 mg/kg / day p.o. for 28 days) on DOX induced (3 mg/kg, IP on days 1, 7, 14, 21, 28) cardiovascular abnormalities in rat heart. DOX...

متن کاملCrataegus monogyna fruit aqueous extract as a protective agent against doxorubicin-induced reproductive toxicity in male rats
Objective: Doxorubicin (DOX) is a broad spectrum chemotherapeutic agent used in the treatment of several malignancies. The use of DOX in clinical chemotherapy has been restricted due to its diverse toxicities, including reproductive toxicity. Crataegus monogyna (C. monogyna) is one of the oldest medicinal plants that have been shown to be cytoprotective because of scavenging free radicals. The ...
متن کاملAntioxidant Effect of Continuous Training with Crocin Consumption on Doxorubicin-Induced Hepatotoxicity in Rats
Introduction: It has been reported that doxorubicin can cause liver damage; however, exercise and crocin consumption can improve antioxidant defense. The aim of the present study was to investigate the antioxidant effect of continuous training with crocin consumption on the liver tissue of doxorubicin-poisoned rats. Methods: In this experimental...
